Disease: Ovarian carcinoma
Derived from tumor tissue of an untreated patient. This cisplatin-resistant cell line has been developed by chronic exposure of the parent cisplatin-sensitive A2780 cell line (C0017002) to increasing concentrations of cisplatin. A2780cis is cross-resistant to melphalan, adriamycin and irradiation. An increased ability to repair DNA damage as well as cytogenetic abnormalities has been observed. In order to retain resistance cisplatinum has to be added to the media for every passage.

AddexBio-formulated RPMI-1640 Medium(C0004-01) + 10% FBS + 1 uM Cisplatin
1:5 using 0.25% trypsin or trypsin/EDTA, 5% CO2; 37°C. Cells will attach slowly after resuscitation and take up to 7 days to reach confluency. Recommendation: recover cells in media without cisplatin. Add after subculture of attached cells.
