The MRC-5 cell line was derived from normal lung tissue of a 14-week-old male fetus in 1966.

Chromosome Frequency Distribution 50 Cells: 2n = 46. This is a normal diploid human cell line with 46,XY karyotype. The modal chromosome number was 46, occurring in 70% of cells. The rate of polyploidy was 3.6%. Both X and Y chromosomes were normal. Cytogenetic instability has been reported in the literature for some cell lines.

Split sub-confluent cultures (70-80%) 1:2 to 1:5 using 0.25% trypsin/EDTA. Culture at 5% CO2, 37°C. Feed cells once every 2 to 3 days.
Complete culture medium supplemented with 5% (v/v) DMSO I
