Origin:
Derivative of L929 selected in azaguanine (8-azaguanine (0.003 mg/mL) and 6-thioguanine (0.1 mM)). The cells are sensitive to HAT selection medium and deficient in adenosine phosphoribosyl transferase (APRT) and hypoxanthine phosphoribosyl transferase (HPRT). A9 may be heterozygous for the ability to synthesise active inosinic acid phosphorylase.

Split sub-confluent cultures (70-80%) at a seed ratio of 1:3 to 1:10, i.e. seeding at 1-5x10,000 cells/cm^2 is recommended, using 0.25% trypsin; 0.05% EDTA. Culture at 5% CO2, 37°C. Renew medium once every 2 to 3 days.
