The STO cell line was derived from a continuous line of SIM mouse embryonic fibroblasts. The population which was selected for 6-thioguanine and ouabain resistance is sensitive to HAT medium and is HPRT negative. Used routinely as feeder layers prepared by irradiation or mitomycinc treatment for maintenance of teratocarcinoma stem cells in the undifferentiated state. The cells have been tested and found negative for ectromelia virus (mousepox).

Split sub-confluent cultures (70-80%) at a seed ratio of 1:3 to 1:6, i.e. seeding at 2-4x10,000 cells/cm^2 is recommended, using 0.25% trypsin; 0.05% EDTA. Culture at 5% CO2, 37°C. Renew medium once every 2 to 3 days.
